Immunological Relationships of Tobacco Isoperoxidases

Abstract
Two cationic isoperoxidases (termed C©ý and C©þ) and two anionic isoperoxidases (termed A©ý and A©þ) have been purified to homogeneity from a tobacco callus culture, Nicotiana tabacum L. var Virginia 115, utilizing ammonium sulfate precipitation, CM-cellulose chromatography, DEAE-cellulose chromatography followed by Sephadex G-100 gel filtration. Antisera against purified isoperoxidases were raised in rabbits, and the quantity of immune IgG which yielded half maximal enzyme inhibition was determined by immunotitration. In Ouchterlony double diffusion experiments, the cationic isoperoxidase C©ý and C©þ revealed an immunological identity, whereas these isoperoxidases had no immunological relationships with anionic isoperoxidase A©ý and A©þ. Only anti A©þ antiserum cross-reacted with horseradish peroxidase preparation, although it did not cross-react with isoperoxidase A©ý. These results suggest an immunological uniqueness of isoperoxidase A©ý among these tobacco isoperoxidases.
